Rv2953 Trans-acting enoyl reductase

Summary

Product Feature Type Start End Strand Length AA Length is TF
Rv2953 Trans-acting enoyl reductase CDS 3305279 3306535 + 1 257 418 FALSE

Rv2953 (Trans-acting enoyl reductase) is predicted to be co-regulated in modules bicluster_0100 with residual 0.55 and bicluster_0126 with residual 0.51.

This regulation is possibly mediated by two de-novo identified cis-regulatory motifs in each module with e-values , 130.00 and 310.00 for bicluster_0100 and 2.80 and 8,500.00 for bicluster_0126 respectively.

These modules are enriched for following go terms: oxidoreductase activity, oxidoreductase activity, acting on NAD(P... glycolysis.

This gene is found to be for growth on cholesterol.

glycolipid biosynthetic process

Details:
The chemical reactions and pathways resulting in the formation of glycolipid, a class of 1,2-di-O-acylglycerols joined at oxygen 3 by a glycosidic linkage to a carbohydrate part (usually a mono-, di- or tri-saccharide).

Actinobacterium-type cell wall biogenesis

Actinobacterium-type cell wall biogenesis

Details:
A cellular process that results in the biosynthesis of constituent macromolecules, assembly, and arrangement of constituent parts of a cell wall of the type found in Actinobacteria. The cell wall is the rigid or semi-rigid envelope lying outside the cell membrane. Actinobacterial cell walls contain characteristic mycolic acids, of which some are covalently linked to the cell wall peptidoglycan and others accumulate at the cell surface.

Quantitative Proteomics Data

t-test p-value Cholesterol/Glycerol Ratio
0.690000 1.33

How essentiality calculations were done?

The relative representation of each mutant was determined by calculating the fold change (sequence reads/insertion in cholesterol divided by sequence reads/insertion in glycerol) for each gene. Statistical significance was determined by t-test. Each insertion site in each replicate sample was treated as a separate data point. The hyperbola used for defining genes specifically required for growth in cholesterol was defined by the formula, y = 3.8/x+0.7. Genes above this line are annotated as required for growth on cholesterol.

TRIP log2 fold abundance change

reports the log2 abundance fold change of each TFI strain, relative to no induction, in absence or presence of drug, averaged across experimental replicates. Also reported are the accompanying z-scores and two-sided t-test p-values for each TFI strain under each condition. Please refer to Ma et al., 2020, Nature Microbiology for more information.
