Diphthine synthase, also known as DPH5; Diphthine synthase, also known as diphthamide biosynthesis S-adenosylmethionine-dependent methyltransferase, participates in the posttranslational modification of a specific histidine residue in elongation factor 2 (EF-2) of eukaryotes and archaea to diphthamide. It catalyzes the trimethylation step in diphthamide biosynthesis. Diphthamide is the target of diphtheria toxin, which ADP-ribosylates diphthamide and inhibits protein synthesis, leading to host cell death.
